(1) Preparation of 5×TBE (tris-boric acid and EDTA) buffer (1000 ml):
Material: Tris 54g, boric acid 27.5g, 0.5mol/LEDTA 20ml
Method: Adjust the pH to 8.0, dilute to 1000 ml, store in a refrigerator at 4 °C, and dilute 10 times when used.
(2) Preparation of loading buffer:
0.25% bromophenol blue, 40% (w/v) sucrose aqueous solution, stored in a refrigerator at 4 °C.
(3) Preparation of ethidium bromide:
Weigh 0.1 g of ethidium bromide, dissolve it in 10 ml of water, prepare a mother liquor with a final concentration of 10 mg/ml, and store in a refrigerator at 4 °C. During the staining, 12.5 μl of the mother liquor was aspirated, and 250 ml of water was added to a final concentration of 0.5 μg/ml, and the mixture was uniformly mixed.
(4) Preparation of 100 times TE buffer:
Material: 1 mol/LTris-HCl (pH 8.0), 100 mmol/LEDTA
Method: Weigh 121.1g of Tris, EDTA-Na237.23g, first use 800ml of double distilled water, stir and dissolve by heating, then adjust the pH to 8.0 with hydrochloric acid (about 20ml of hydrochloric acid), and then make up to 1000ml.
(5) Preparation of 100 times electrophoresis buffer:
Material: 4mol/LTris-HCl (pH8..0), 2mol/L sodium acetate, 200mmol/LEDTA
Method: Weigh 242.2g of Tris, 82.03g of anhydrous sodium acetate, 237.23g of EDTA-Na, firstly stir and dissolve with 400ml of double distilled water, then adjust the pH to 8.0 with glacial acetic acid (about 50ml of glacial acetic acid), then set Allow up to 500ml. Dilute 100 times when used.
